what i did (and usually do when i start a modification of some type) is find out the structure. I have to admin I'm a big fan of Misa's texturepack, but even a bigger fan of using it to understand the resource pack.
download misa's texture pack, then open the zip file and learn the folder structure, and the file names. Then you can make you own separate zip with your own custom png's in it. easy peasy

try searching in blogs: HOW TO MAKE RESOUCE PACKS, if you want to edit sound, i have a blog on the subject, to redo textures, get the program(s) that jetra mentioned before, then find the textures in the Minecraft.jar, you then redo the textures you want, also, if you would like, you could download a texture pack, and redo the textures in that (just don't upload it here, it'd be a personal use pack)

First, you need a proper program to make textures with. I would suggest either GIMP or paint.NET, both are good at simple texture designs and offer a variety of colors.
