Can't you just put a random block where the diamond block was and then run away?

Yes thats the problem i also need an idea how i could force the player to go in with no other blocks...

You could have a bed that they can sleep in, and make the door to the diamond room be locked until they go into go into a killing machine. If they exit the room or go back into the killing machine to recollect their items, they step on a pressure plate which locks the door again.

couldnt you make a piston push out where the diamond block used to be so the person cant put it back?
