It's a very complicated process, because all of the blocks (except for cactus, Pumpkin and Jack O' Lantern) WILL need to have Seamless textures - Whichever will be your style: Comic, realistic, simple, all of them will need to be seamless. It's hard to find the one you need, but when you do it's always a pleasure
Also, you can turn a texture seamless in Photoshop. There's some tutorials in Youtube, you should find it there

Follow these steps to create a pack:
1. Get GIMP or Paint.NET
2. Get 7zip or WinRar.
3. Hit Start, type %appdata% and hit enter.
4. Go into .minecraft > bin in the window that comes up.
5. Right-click the file called "minecraft" within that folder, and select 7zip > Open Archive (or WinRar)
6. Find and take out folders such as: terrain, gui, mobs, textures, pack.txt and a few more.
7. Edit with GIMP/Paint.NET.
8. Select all edited files, and click Send To > Compressed (Zipped) folder.
That's it!

That's a very broad question you have there. It's best to observe other textures and experiment. Texturing things is not a skill you gain over night.

Go YouTube how to, it is really complex editing the textures. Sorry for the lame advice.
