Make a portal out of glowstone, exactly like you would with obsidian with the nether portal. Then instead of right clicking on it with flint and steel, get a bucket of water and right click the inside of it. Then there ya go

you find ender portals in newly generated strongholds. you need 12 eyes of ender to activate them, eyes of ender are made from blaze power and ender pearls. eyes of ender also float in the direction of the closest stronghold when thrown
