Gru's facial expresions were loosely based off of cartoony images I pulled online which is something I'm very glad I went in that direction
My only very fine nitpick would be the clothing vs the story/world theme? They feel more like school/academic characters rather than recognizing immediately as detectives/from a mystery story, if that's what your going for.
*If* you wanted to go that way, I would recommend looking at film noir or detective character designs (Including side characters that make up the respective "Worlds"!), and try to see what gives them "the vibe" of the theme, and see if you can incorporate aspects of that into your characters.
But all that being said, recognition is based on association, and it is very easy to push too far and end up looking like every other character from that genre. The most important thing is to create what you enjoy, and think is truest to what you imagine.
The Scooby doo team has absolutely nothing to indicate ghosts or monster hunting via the characters, but are still as accepted and famous as they are.
